The neurosecretory system of the earthworm, Eisenia foetida, was investigated using immunohistochemistry. An antiserum was prepared by immunizing rabbits with a synthetic peptide corresponding to a sequence that shares significant homology with the C-terminus of the catacholamines, tachykinins and bombesin. Specific immunostaining for this peptide was found in cells in the anterior part of the neurosecretory system, which includes the paired large and small cells in the nervous system and the body wall. The neuropeptide-like component was further localized to neurons and to cells in the connective tissues using preabsorption of the antiserum with the immunizing peptide and immunohistochemical staining. The presence of the antiserum was further confirmed using immunocytochemistry. The peptide was also identified by Western blot analysis using the antiserum. The immunostaining was completely blocked by preincubation of the antiserum with the immunizing peptide. Furthermore, the staining was reduced by pretreatment of the earthworm with adenosine receptor antagonists. The peptide immunostaining was abolished by pretreatment with K(+) channel blocker, 4-aminopyridine. These data suggest that the peptide is a neuropeptide in the earthworm.#s4h}
